چکیده
Abstract BACKGROUND Leptomeningeal disease (LMD) occurs in approximately 5% of patients with breast cancer and has a median survival 2-4 months. We found loss the anti-HER2 anti-HER3 CD4 Th1 immune responses patients. In pre-clinical clinical trials, administration class II HER2 peptide-pulsed dendritic cell vaccine (HER2-DCV) partially restores pathologic complete HER2+ Here, we examined intrathecal (IT) delivery HER2/HER3-DCV cancer-associated LMD immunocompetent animal models. METHODS Luciferase-labeled TUBO cells were injected into cisterna magna BALB/c mice to produce LMD. used our Murine Ommaya (mimics an reservoir patients) for IT DCVs CSF. RESULTS AND DISCUSSION cancer-LMD randomized following groups: 1) HER2-DCV 2) HER3-DCV 3) IT. The untreated (control) group was 15 days. All groups given DCV prolonged (p< 0.001). Interestingly, HER2-/HER3-DCV able rescue (71% 28% triple negative cancer-LMD) showed tumor regression. Some surviving subsequent rechallenge. CSF, evidence CD4+ CD8+ T-cells infiltration, robust IFN-g IL18 response upon treatment. CONCLUSIONS Our preclinical data supported trial (submitted)

Design of a Novel Recombinant Multi-Epitope Vaccine against Triple-Negative Breast Cancer
Background: Triple-negative breast cancer (TNBC) is determined by the absence of ERBB2, estrogen and progesterone receptors’ expression. Cancer vaccines, as the novel immunotherapy strategies, have emerged as promising tools for treating the advanced stage of TNBC. The aim of this study was to evaluate Carcinoembryonic antigen (CEA), Metadherin (MTDH), and Mucin 1 (MUC-1) proteins as vaccine ...
متن کاملMammographic findings in different breast cancer subtypes (luminal, Her2 positive, triple negative)
Background: Prognosis and management of breast cancer are defined by different variables including histological type, grading, clinical stage, Her2+, estrogen and progesterone receptor conditions. Generally, mammography is one of the most important imaging which is done in breast cancer patients. The aim of this research was to evaluate different mammographic patterns in different breast cancer...
متن کاملDelphinidin Inhibits HER2 and Erk1/2 Signaling and Suppresses Growth of HER2-Overexpressing and Triple Negative Breast Cancer Cell Lines
Delphinidin is a polyphenolic compound found in many brightly colored fruits and vegetables. Delphinidin is also the major bioactive component found in many dietary supplements that are currently consumed as complementary cancer medicine including pomegranate extract. The purpose of the current study was to determine the in vitro biological effects of delphinidin on established breast cancer ce...
متن کاملDipalmitoylphosphatidic acid inhibits tumor growth in triple-negative breast cancer
Triple-negative breast cancer (TNBC) is a subtype of breast cancer with a poor prognosis, accounting for approximately 12-24% of breast cancer cases. Accumulating evidence has indicated that there is no effective targeted therapy available for TNBC. Dipalmitoylphosphatidic acid (DPPA) is a bioactive phospholipid. However, the function of DPPA in the growth of TNBC has not yet been studied. In t...
متن کاملAssociation of HDAC8 Expression with Pathological Findings in Triple Negative and Non-Triple Negative Breast Cancer: Implications for Diagnosis
Background: Previous data have shown the tumorigenicity roles of HDAC8 in breast cancer. More recently, the oncogenic effects of this molecule have been revealed in TNBC. The present study aimed to determine the diagnostic value of HDAC8 for the differentiation of TNBC from nTNBC tumors. Methods: A total of 50 cancerous and normal adjacent tumor specimens were obtained, and the clinical and pat...
